Method: Immunocytochemistry Unit: Staining index Protocol: Cytocentrifuge preparations of tumor cell lines were air-dried and fixed in acetone. Slides were incubated with LMR-12 or isotype matched rat Ig, then with biotinylated rabbit anti-rat IgG or anti-mouse IgG and streptavidin-HRP. Color development was evaluated on coded slides to avoid bias in scoring. A semiquantitative 'staining index' was calculated as the product of the fraction of positive cells and the average staining intensity estimated on a scale from 0 (negative) to 3 (very strong positive). At least three tests for each cell line were used for calculation of the average staining index. |
