Method: PCR Slot Blot Unit: RNA levels (zero values replaced with 3000 counts for kinases and 2000 counts for phosphatases) Protocol: RNA from each cell line was PCR amplified using selected pools of degenerate primers to generate 32P labelled probes. Probes were hybridized to slot blots containing equimolar amounts of known kinases and phosphatases. Results are presented as background adjusted counts for all DNAs hybridized. Zero values were replaced with 3000 counts for kinases and 2000 counts for phosphatases, to permit the generation of log-average plots of the data. Northern blot analysis, quantitative PCR, and RNAse protection assays were performed on selected kinases to confirm the results obtained using this protocol. |
